The track’s start and finish lines are strategically positioned on Ayala Avenue, one of Makati’s main thoroughfares. From there, the course proceeds clockwise through the central business district, navigating prominent commercial hubs such as Glorietta and Greenbelt. This section features a combination of tight corners and short straights, demanding precision and technical skill from drivers.

After negotiating these complex urban sectors, the circuit opens up on Arnaiz Avenue, allowing drivers to accelerate and gain speed before a right-hand turn onto Amorsolo Avenue. The route then weaves through Legazpi Village, a mixed-use area known for its modern architecture and green spaces, adding a scenic dimension to the race.

At 4.732 kilometers (approximately 2.94 miles) in length, the Makati Circuit is marginally longer than the Circuit de Barcelona-Catalunya, which measures 4.655 kilometers.
